New Super Mario Bros. has got to be my favourite DS title. Super Mario 64 DS is great too (except the broken controls, but with excellent replayability).

A lot of DSiWare games are awesome but I'm guessing you have a Phat or Lite.
Less known Games? Both RPGs: The World Ends With You (Great Unique Battling; Great Plot) & Dragon Quest V: The Hand Of The Heavenly Bride (Addictive; Great Plot; Absolutely MASSIVE!)
@Bobpie TWEWY is very well-known. It's almost at the level of Kingdom Hearts on the popularity scale. The SMT series is a truly lesser-known set of games which blow TWEWY out of the water imo.
